Reactions - Sometimes the humor is not in the gag line or the situation as much as it is in the reaction of the characters. Comedian Jack Benny was great at using this formula. One of his supporting players would tell a joke and he would just give you this reaction stare. Usually you laughed more at the expression on Jack Benny's face than you did at the gag.

Look at the cartoon on the left. The real humor in this cartoon, in my opinion, is in the reaction on the man's face.
Misunderstanding - When one character understands one thing about a situation and another character interprets it entirely different, it can make for some funny gags. Take the cartoon on the right as an example. This man's boss meant something entirely different than the happy employee understood.
